 High Potency (Hi-Po) SYBR Green C-to-C  Kit is developed to quantitatively analyze cellular genomic DNA-free RNA samples obtained directly from cultured cells for reverse transcription and quantitative PCR (RT-qPCR) applications without RNA purification process.

In Hi-Po SYBR Green C-to-C  System, preparation of RNA samples for RT-qPCR requires only two steps of pipetting to add reagents, gentle hand-rotation/shaking/tumbling of the sample container, and incubation of the samples at room temperature for 5-7 minutes. There is no repetitive pipetting to mix solutions, sample transfers, centrifuging, heating or cooling. Characterized by this extraordinary ease and speed, C-to-C  System is an exceptionally robust system when a lot of samples (such as in 96/384-well plates) have to be processed simultaneously.

Traditionally, the first step in mRNA/gene expression examination involves purification of RNA from experimental samples. The conventional RNA isolation is fairly time consuming, often requiring >30 minutes of hands-on sample manipulation. With fewer steps and less hands-on manipulation, C-to-C  System can process RNA samples from 10-100,000 cells ready for RT-qPCR without RNA purification, substantially expediting and simplifying gene expression analysis of cells. For example, when using C-to-C  System, it takes approximately 10/15 minutes to prepare 96/384 RNA samples ready for the downstream RT-qPCR from cells cultured in a 96/384-well plate including genomic DNA removal if necessary.

More importantly, in the absence of sample transfers during RNA sample preparation,  Hi-Po SYBR Green C-to-C System minimizes RNA loss and exhibits sensitivity and specificity comparable to the performance from column-purified RNA in RT-qPCR. Due to the simple cell lysis step – 5-7 minutes at room temperature without sample transfers, C-to-C  System can be conveniently automated with robotic platforms for high-throughput processing of 96/384-well plates. The procedure is also economically and environmentally friendly due to the greatly minimized workflow procedure from our C-to-C  System.
