This cryopreservation medium is devoid of serum, proteinaceous constituents, and dimethyl sulfoxide (DMSO), offering enhanced safety and usability. Its composition is explicitly defined, ensuring reproducibility and reliability. It facilitates robust cell recovery rates while preserving cellular functionality, rendering it applicable across a broad spectrum of cell types, including the establishment of cell repositories, long-term storage of research cell lines, and preservation of various mammalian cell populations such as mesenchymal stem cells, mononuclear cells, and immune cells.
In comparison to conventional serum-based cryopreservation media, this formulation mitigates the potential risk of introducing exogenous viruses associated with serum. Moreover, it addresses the limitations of existing serum-free, DMSO-free cryopreservation solutions by circumventing the presence of exogenous proteins. Additionally, it exhibits an extended shelf life. Even after 12 months of storage, cells cryopreserved using this medium maintain a recovery rate exceeding 90%.
Protocol:
1. Harvest the cells and centrifuge according to cell type.
2. Remove the supernatant carefully.
3. Add cryopreservation media. The volume of media used should be calculated according to cell type and desired concentration.
The concentration recommended is 0.5~5*10^6 cells/ml for adherent cells, or 0.1~2*10^7 cells/ml for suspension cells.
4. Gently resuspend the cells by pipetting up and down for 4-5 times to spread cells evenly in the media.
5. Aliquot into cryogenic storage vials. Label accordingly.
6. Store in -80℃ freezer overnight, then move to liquid nitrogen tank for long term storage. Use of a programmed cryopreservation container is optional.
